Cleaning up 404 to 301 on Website

Hi, I can work on creating the spreadsheet with the comparative url's and will redirect the 404 urls to new ones via 301 redirect. For the redirects do i have to enter the url's in the cms or have to add them in the .htaccess file, please let me know.
